一、荧光染料（粉末形式，特别是对氧敏感探针）配制储存液的注意事项
|
|
1）荧光探针在固体（粉末）状态很稳定，按照说明书要求温度来保存，效期内使用即可。
2）荧光探针用有机溶剂比如：DMSO，溶解配制成储存液之后，一般来说，放到-20℃保存（2-3个月内使用，甚至更短，具体可咨询）；放到-80℃保存（6个月内使用，具体可咨询）。前提是，使用的有机溶剂必须是高质量且无水的，特别是DMSO，必须是新鲜开封。
3）配制的储存液请务必用密封性好且螺旋盖的低容量冻存管保存（不可用EP管），至少按照5-10ul/管来分装，避光冻存。
4）对于某些特殊化合物（对空气敏感或存在不稳定结构），可能保存周期特别短，甚至只能当天使用。这些化合物说明书上会有说明。

二、荧光染料（以溶于有机溶剂的储存液形式提供）的注意事项
|
|
1） 以溶于有机溶剂的储存液形式的荧光探针相对来说是比较稳定的化合物；但收到这类产品，也需用户根据单次用量（5-10ul/管来分装），-20℃以下密封避光保存，减少反复冻融次数。
2） 请务必用密封性好且螺旋盖的低容量冻存管保存（不可用EP管）。务必避光。

产品描述：
色霉素A3（Chromomycin
A3，CMA3，CAS：7059-24-7），同义名Aburamycin b，由灰色链霉菌（Streptomyces griseus）代谢纯化所得的一种蒽醌抗生素，具有抗菌、抗真菌和抗肿瘤活性。常用作一种荧光DNA染料，非常适合用来筛查精子染色质存在的精蛋白缺乏（protamine
deficiency）。在二价金属离子存在的条件下，色霉素A3（CMA3）通过与DNA特异性结合与可逆作用来阻断大分子合成。另外，通过结合到DNA小沟，色霉素A3有效调控与DNA促旋酶的竞争性抑制，从而显著影响拓扑异构酶II活性。CMA3对非小细胞性肺癌和宫颈癌呈现体外细胞毒性，作用于HCC44，A549，ME180和 HeLa细胞的IC50s分别是1，42，60和40nM。CMA3还能通过改善Sp1和Sp3转录因子结合来抑制氧化应激和DNA损伤诱导的神经损伤。具体内活性。

spermatozoa were evaluated under a Nikonfluorescent microscope.

实验方法（Assay）：The spermatozoa were
washed again, fixed in methanol/glacial acetic acid (3:1) at 4℃ for 5 min and
then spread on slides. For CMA3 staining, each slide was treated for 20 min
with 100µl of CMA3 solution (0.25 mg/ml in McIlvaine buffer, pH 7.0,
containing 10 mM MgCl2). The slides were then rinsed in buffer and
mounted with buffered glycerol. Microscopic analysis of the slides was
performed on a fluorescent Zeiss axioplane microscope with the appropriate
filters.

dissolved in ethanol as 1:1000 stock solution.
实验方法（Assay）：PC3 cells were seeded
at indicated densities in 12-well plates in triplicates and treated with
DMSO, atorvastatin (10 μM), cyclohexamide (10 μg/ml) and chromomycin A3 (5
μM) or atorvastatin plus cyclohexamide or chromomycin A3. The cells were
cultured for 2, 3 and 4 d, trypsinized, and counted by the cell counter
Cellometer Auto T4.
