产品描述 天狼星红染色(Picro Sirius Red Stain)是一种简单和灵敏的方法用于鉴定组织切片中的胶原纤维网络,通过形态成像分析来定性或定量鉴别退行性病变和遗传性或后天性疾病内胶原网络的异常变化。天狼星红是一种强且线性的阴离子染料,包含6个磺酸基,能够与阳离子胶原纤维牢牢结合。偏振光检测下,胶原纤维有正的单轴双折射光属性,当与天狼星红染料结合后,胶原的天然双折射属性得以增强,从而能区分两型胶原纤维。
本天狼星红染色试剂盒(Picro Sirius Red Stain Kit)主要由天狼星红染色液、Mayer苏木素染色液组成,用于胶原纤维I和胶原纤维III的染色。在偏振光下,通过对胶原纤维的双折射光来区分I型胶原和III型胶原。也能在光学显微镜下观察胶原染色。本染色液稳定,不易褪色。且操作简单。

注意事项 1) 为使偏振光镜下显色清晰,切片厚度以6~7μm为宜。 2) 复染细胞核宜用Mayer’s苏木素染色液,其不影响I型胶原和III型胶原的数量和双折射强度。 3) 为了您的安全和健康,请穿实验服并戴一次性手套操作。
操作步骤 一、 需要自行准备的材料 1) 10%福尔马林固定液(比如:MM1519-500ML) 2) 中性树胶封片剂(比如:MM1503-100ML) 3) 普通光学显微镜或偏振光显微镜
二、染色步骤 1) 组织固定于10%福尔马林固定液,常规脱水包埋。 2) 切片厚度6~7μm为宜,以按常规方法脱蜡至水。 3) 天狼星红染色液滴染1h。 4) 流水稍微冲洗,去掉切片表面染色液。 5) Mayer’s苏木素染色液染细胞核8~10min。 6) 流水冲洗10min。 7) 常规脱水透明、中性树胶封固。 8) 普通光学显微镜或偏振光显微镜下观察。

The liver tissue was immediately fixed in a 10% formaldehyde solution and gradient ethanol for dehydration treatment, then embedded in paraffin to make 5 μm thick tissue sections. Before pathological staining, the slices were placed in an oven at 60 ℃ for 2 h, then dewaxed with xylene and rehydrated with gradient ethanol. The nuclei were then stained with hematoxylin for 1 h, dehydrated, and sealed with clear gum. Two types of collagen fibers could be seen under the polarised light microscope. Type I collagen fibers were closely packed and showed strong birefringence, with yellow or red fibers. Type III collagen showed weak birefringence, with fine green fibers. It carried out the specific procedures of this experiment according to the instructions of Shanghai MKbio Biological Technology Co., LTD. Sirius red Staining Kit (cat no. MM1004). The experimental results were measured by Image-Pro Plus 6.0 (IPP 6.0), and the results were presented in the form of area proportion.

For Picrosirius Red staining, the Picro Sirius Red Stain Kit (MaoKang Bio, China) was used. Briefly, cell sheet sections were deparaffinized and hydrated with distilled water. The tissue section was adequately covered in Picrosirius Red Solution.

Histopathological changes analysis Hematoxylin and eosin (H&E), Masson and Sirius red staining were performed to evaluate the histopathological changes in vivo44. Briefly, heart tissue was sliced into 8-µm-thickness sections and fixed with 4% paraformaldehyde at room temperature.
