产品描述
番茄凝集素(Tomato lectin,Lycopersicon Esculentum Lectin(LEL)),是一种非常稳定的单亚基糖蛋白,约含有等量的蛋白质和碳水化合物。据报道其糖类部分由85%阿拉伯糖和15%半乳糖组成,在溶液中可能形成多聚体聚合物。LEL无血型特异性,但能亲和N-乙酰-β-D-葡糖胺寡聚体。LEL能够很好的结合内皮细胞,用来观察和评价微血管系统。LEL能够抑制菜豆来源植物血凝素(PMA)的促有丝分裂活性。
本品为FITC标记的番茄凝集素(来源于Lycopersicon esculentum),Ex/Em=495/515nm,通过血管内灌注法或直接染色组织切片能快速且有效的检测和可视化啮齿类动物的血管和小胶质细胞。

体内研究(Lectin Perfusion):
动物模型(Animal Model):Xenografts were established in 4- to 6-week-old female NCR
nude mice by intrarenal injection of 106 SK-NEP-1 cultured human Wilms tumor
cells and allowed to grow. After 5 weeks, large tumors were palpable in all
mice, and a cohort was randomly selected to provide day-0 controls.
灌注方法(Lectin
perfusion):Selected
mice at each time point underwent intravascular injection of fluorescein- labeled
Lycopersicon esculentum lectin (100 μg in 100 μl of saline) into the left
ventricle. The vasculature was fixed by infusion of 1% paraformaldehyde (pH
7.4) in PBS and then washed by perfusion of PBS.

动物模型(Animal Model):Tumor-bearing RIP-Tag2 transgenic mice34 (C57BL/6 background,
10 to 12 weeks of age).
灌注方法(Lectin
perfusion):In mice used
for immunohistochemistry, blood flow and patency of individual tumor vessels
were assessed by injection of 100 μg of fluorescein isothiocyanate-labeled
Lycopersicon esculentum lectin in 0.9% NaCl (100 μl into femoral vein). Two
minutes later the chest was opened rapidly, and the vasculature was perfused
for 2 minutes at a pressure of 120 mmHg with fixative [1% paraformaldehyde in
phosphate-buffered saline (PBS), pH 7.4] from an 18-gauge cannula inserted
into the aorta via an incision in the left ventricle. Blood and fixative
exited through an opening in the right atrium. Tissues were removed, immersed
in fixative for 1 hour at 4°C, and then processed for immunohistochemistry.

实验方法(Assay):Take 25 μl of 2mg/ml Lycopersicon esculentum (tomato) lectin
FITC conjugate stock solution into 2.5ml r-ACSF to give 20 μg/ml
concentration. Pipette the dye over the slices as before, leaving the slices
incubating for a period of 45 minutes.
