芝加哥天蓝6B(Chicago Sky Blue 6B, CSB6B),又称为滂胺天蓝(Pontamine sky blue),是一种染料和谷氨酸盐衍生物,具广泛的生物活性。芝加哥天蓝6B(CSB6B)是囊泡谷氨酸转运体(vGLUTs)的竞争性抑制剂,CSB6B(0.1 mM)抑制大鼠脑部突触囊泡的谷氨酸摄取(Ki=0.19µM),但不抑制突触体介导的谷氨酸摄取。CSB6B也是钙/钙调素依赖性蛋白激酶磷酸酶(CaMKP)和其核异构型CaMKP-N 的抑制剂(IC50s分别是4.1和1µM)。CSB6B(17µM)抑制钠离子-牛磺胆酸共转运多肽(NTCP, IC50= 7.1µM),降低HepaRG细胞感染乙型肝炎病毒(HBV)和丁型肝炎病毒(HDV)。脑室注射CSB6B(0.5µg)抑制小鼠炎性疼痛,减低乙酸诱导的扭动和福尔马林II期的伤害行为,对自发运动无影响。CSB6B还能被用作复染剂,降低免疫荧光组织化学制备中的背景自发荧光。

配制方法(Preparation):Chicago Sky Blue 6B (CSB) was dissolved in DDW to 10 μM for all in vitro cardiac cell culture experiments, and 50 μM for BMDM experiments.
实验方法(Assay):Bone marrow cells were isolated and plated in macrophage differentiation medium. After 6 days of culture, the medium was aspirated and replaced with fresh macrophage differentiation medium with the addition of 10 ng/mL LPS. After 24 hours the LPS-containing medium was aspirated, cells were washed twice with PBS, andwere treated with 50 μM CSB in macrophage differentiation medium. Twenty-four hours later the cells were washed and RNA was purified.
体内研究:
动物模型(Animal Model):12-week-old Hsd:ICR female mice (Envigo RMS)
给药剂量(Dosages):In vivo: CSB was diluted in PBS. Injection by i.m.: 50 μL of 100 μM stock solution, which is 0.005 μmoles (5 μg). Injections by i.v.: 100 μL of 100 μM stock solution, which is 0.01 μmoles (10 μg).
实验方法(Assay):Mice were sacrificed 21 days after MI following 7 injections of CSB. The first injection was 50 μL of 100 μM CSB and was injected i.m. into the myocardium. Six additional injections were 100 μL of 100 μM CSB and were injected i.v. into the tail vein. The last i.v. injection was given 14 days after MI. PBS-treated mice were examined for comparison. A total of 6 CSB-treated and 5 PBS-treated mice were examined.

动物模型(Animal Model):Female C57BL/6 mice (6–10 weeks of age)
药物配制(Preparation):1% (w/v) Chicago Sky Blue 6B in PBS
实验方法(Assay):To visualise implantation sites,mice were injected intravenously with 0.1ml of 1% (w/v) Chicago Sky Blue 6B in PBS under anaesthetic and sacrificed by CO2 asphyxiation 10min later. Individual decidua were removed from the uterus and shaken in 25μl of formamide for 2 days at room temperature (RT), protected from light to extract the dye. After clearing by centrifugation (16,000×g for 5min), dye content in formamide extracts was quantitated spectrophotometrically using an ND1000 Nanodrop at 618nm relative to a standard curve of Chicago Sky Blue dye dissolved in formamide.
